چکیده

PURPOSE To investigate the in vitro effects of gentamicin sulfate, vancomycin hydrochloride, sodium cefazolin and ceftriaxone on glucose 6-phosphate dehydrogenase enzyme (G6PD) purified from sheep lenses. METHODS G6PD was purified from sheep lenses with a yield of 66.8% and a specific activity of 7.8 U/mg proteins, and 10,400-fold using ammonium sulfate fractionation and 2,5-ADP Sepharose 4B affinity gel. The enzyme activity was determined by Beutlers method. RESULTS Gentamicin sulfate and vancomycin hydrochloride strongly inhibited the enzyme in vitro. The concentrations causing 50% inhibition (IC50) were 15.34, and 8.0 mM, respectively. Conversely, cefazolin sodium strongly activated this enzyme, and ceftriaxone caused milder activation. CONCLUSIONS If a patient with G6PD deficiency requires gentamicin sulfate or vancomycin hydrochloride, routine ophthalmic did not inhibit this enzyme. Postmortem studies are now needed to investigate the activity of G6PD and how it is affected by these antibiotics. (Eur J Ophthalmol 2003; 13: 155-61).

Protective Effect of Quercetin on Oxidative Stress in Glucose-6-Phosphate Dehydrogenase-Deficient Erythrocytes in Vitro

Glucose-6-phosphate dehydrogenase (G6PD) deficient subjects are vulnerable to oxidative stress. Quercetin, a flavonoids, has been employed as a potent oxygen-free radical scavenger in order to assess the protective effects of quercetin against H2O2-induced oxidative damage in G6PD-deficient and normal human erythrocytes. Erythrocytes of G6PD-deficient (n = 10) and normal (n = 10) subjects were ...

Effects of some antibiotics on glucose 6-phosphate dehydrogenase in sheep liver

In vitro effects of penicillin, sulbactam, cefazolin, and amikacine on the activity of the enzyme glucose6-phosphate dehydrogenase in sheep liver were investigated. Glucose 6-phosphate dehydrogenase was purified from sheep liver, using a simple and rapid method. e purification consisted of two steps, preparation of homogenate and 2’, 5’-ADP Sepharose 4B affinity chromatography. As a result of ...

P-90: Effect of Phosphodiesterase Type3 Inhibitor, Cilostamide, on The Developmental Competence of Ovine OocytesIsolated by Glucose 6-Phosphate Dehydrogenase Activity

Background: The developmental competence of oocytes matured in vitro (IVM) is yet far below than in vivo counterparts. Recent studies suggest that the asynchrony between nuclear/cytoplasmic maturation and the initial low/heterogeneous quality of oocytes are the most important factors affecting IVM success. We investigated whether selection of growing oocytes (based on their glucose 6- phosphate...

The Effects of Hypo- and Hyperthyroidism on Glucose 6-Phosphate Dehydrogenase Activities in Regions of Rat Brain

The variations of glucose 6-phosphate dehydrogenase (G6PD) activity in different brain regions of normal, hypo- and hyperthyroid rats were investigated. Hypo- and hyperthyrodism were experimentaly induced by administration of methimazol and liothyronine, respectively. In normal rats, midbrain had the minimum (70 mU/mg) and cerebral cortex the maximum (349 mU/mg) G6PD activity. Hyperthyrodism in...
